INFORMATION: Interleukin-2 (IL-2) is a 133 amino acid glycoprotein with one intramolecular disulfide bond and variable glycosylation (1). It is secreted by activated T cells and induces proliferation and maturation of activated T cells, natural killer cells, and lymphokine activated killer cells. IL-2 also stimulates proliferation of antibody-producing B cells, activates neutrophilis, and induces mononuclear cells to secrete gamma interferon and tumor necrosis factors alpha and beta (1-4). Moreover, recent studies have shown that IL-2 is required foractivation induced apoptosis, an important hemeostatic mechanism in the immune system, which involve in the maintenance of peripheral tolerance to self-antigens (5, 6). A mouse IL-2/Fc fusion protein is made by genetically fusing IL-2 to Fcg2a. This fusion protein possesses both the biological functions of the IL-2 moiety and a prolonged circulating half-life determined by the Fc domain. Mutations to the complement (C1q) and FcgR I binding sites of the Fcg2a fragment render IL-2/Fc incapable to direct antibody directed cytotoxicity (ADCC) and complement directed cytotoxicity (CDC) (7). This reagent is used successfully in basic and applied research (8 -10).

INFORMATION:IL-15 is a 14- to 15-kDa member of the 4 alpha-helix bundle family of cytokines that possess T cell growth-factor activity. INFORMATION:IL-15 is a 14- to 15-kDa member of the 4 alpha-helix bundle family of cytokines that possess T cell growth-factor activity. In contrast to IL-2, a T cell product, IL-15 mRNA is expressed by a wide varity of cells, including macrophages, B cells, thymic, activated vascular endothelial cells, and bone marrow stromal cells, as well as tissues such as liver, heart, spleen, lung, and skeletal muscle. IL-15 exerts overlapping activities with IL-2 due to their shared B and y-chain receptor components. A receptor site-specific IL-15 antagonist, mutant human IL-15/mouse Fc, is made by mutating glutamine residues within the C terminus of IL-15 to aspartic acid and genetically fusing to mouse Fcy2a. This mutant IL-15 fusion protein specifically binds to the IL-15R, competitively inhibits IL-15-triggered cell proliferation, does not activities the STAT-signaling pathway, and possesses a prolonged circulating half-life determined by the Fc domain. Since the Fcy2a murine isotype is able to bind effectively to cells expressing the high affinity FcyR I receptor and possesses a complement (C1q) binding domain, this is able to facilitate antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The cytolytic mutant IL/Fc fusion protein is potentially able to target the cells bound by IL-15 moieties.

INFORMATION: The dual probe CTLA4/Fc ELISA Kit is a quantitative "sandwich" enzyme immunoassay. A rat IgG antibody specific for mouse CTLA4 has been coated onto the microtiter plates provided in the kit. Test samples or CTLA4/Fc standards are added to the mocrotiter wells and any CTLA4 present binds to the pre-coated IgG antibody. After washing away unbound sample components, an enzyme-linked rabbit IgG specific for mouse IgG2a Fc is added to the wells. A sandwich is thus formed with the CTLA4/Fc bound by the coated anti-CTLA4 antibody and detecting anti-mouse IgG 2a Fc antibody. Thus the dual probe ELISA ensures this assay specific for mouse CTLA4/Fc fusion protein but neither CTLA4 nor IgG2a. Excess enzyme-conjugated IgG is removed by washing and a substrate solution is added to the wells. Color develops in proportion to the amount of CTLA4/Fc present in the sandwich.
In addition to the samples tested, a Standard Curve is prepared using known concentrations of the CTLA4/Fc standard provided in the kit. The optical density of the test samples is compared with the Standard Curve and the CTLA4/Fc concentration in the sample is determined.
Dual probe CTLA4/Fc ELISA ensures this assay specific for mouse CTLA4/Fc fusion protein but neither CTLA4 nor IgG2a.

INFORMATION:The 18d3 monoclonal antibody reacts with mouse CD94, a 70 kDa type II transmembrane glycoprotein. CD94 belongs to the C-type lectin superfamily and is present as a heterodimer with NKG2 on the surface. CD94 is expressed by NK cells and NKT cells, and plays an important role in adhesion and activation of NK cell lineage. CD94/NKG2 is expressed on Th1 but not Th2 cells and costimulates Th1 effector functions. DBA/2J mice are naturally CD94-deficient and do not express cell-surface CD94/NKG2A receptors, even on neonatal NK cells.

INFORMATION:Tim-3, a type I transmembrane protein, contains an immunoglobulin and a mucin-like domain in its extracellular portion and a tyrosine phosphorylation motif in its cytoplasmic portion. INFORMATION:Cytolytic T lymphocyte-associated antigen, CTLA-4 or CD152, is a cell surface glycoprotein that is transiently expressed at low levels on activated T cells. CD152 is a high affinity receptor for the costimulatory molecules CD80 (B7-1) and CD86 (B7-2) expressed on antigen presenting cells (APC). This interaction appears to deliver a negative regulatory signal to the T cell. While CD28 binds to CD80 and CD86 with lower affinity and deliver positive signal fro T cell activation and proliferation. CD28 and CD152 play important roles in regulating the magnitude and nature of T cell mediated immune response. A monoclonal antibody (20A) has been extensively characterized as being capable of blocking human B7-1/2 binding to CTLA-4.
