IL-21R (mouse):Fc (mouse) (rec.) (non-lytic)

Product Data

Synonyms :

Interleukin-21 Receptor; CD360; NILR

Properties

Host :

CHO cell

Sequence :

The extracellular domain of mouse IL-21R (AAL82636.1)(Cys20-Trp234) is fused to the N-terminus of the Fc portion of a mutant mouse IgG2a.

Molecular Mass:

51 KDa (monomer)

Non-lytic:

Acts as a long lasting fusion protein which only binds to the receptor.

Mutations to the complement (C1q) and FcgR I binding sites

of the IgGs Fc fragment render the fusion proteins incapable of antibody

directed cytotoxicity (ADCC) and complement directed cytotoxicity (CDC).

Purity :

>98%by SDS-PAGE under reducing conditions.

Endotoxin Content :

<60EU/mg as determined by LAL test.

Activity:

Measured by its ability to inhibit IL-21-dependent proliferation of N1186 human T cells.

Shipping and Handling

Formulation :

Lyophilized from 0.2μm-filtered solution in PBS.

Reconstitution :

Reconstitute at 100μg/ml in sterile PBS.

Use / Stability :

Stable for at least 1 year after receipt when stored

at -20°C. Working aliquots are stable for up to 3 months

when stored at -20°C.

                        Avoid freeze/thaw cycles.

Description

Interleukin-21 receptor (IL-21R) is a type I transmembrane glycoprotein within the class I cytokine receptor family, type 4 subfamily. Complex formation between IL-21R and the common γ chain (γc) is required for signaling. IL-21R is expressed mainly on B cells (highest on mature, activated, follicular and germinal center B cells), NK cells and activated T cells, but is also found on dendritic cells, alternatively activated macrophages, intestinal lamina propria fibroblasts and epithelial cells and keratinocytes. B cell IL-21R engagement induces Blimp1 (which mediates plasma cell differentiation) and is important for memory responses. IL-21R engagement on mouse NK cells enhances their cytotoxic activity and IFN-γ production. IL-21R engagement on CD8+ T cells aids control of viral infection and tumor growth; IL-21R is also necessary for sufficient numbers of regulatory T cells to combat chronic inflammation. IL-21R expression is

often upregulated in allergic skin inflammation, systemic lupus erythematosus and diffuse large B cell lymphoma (DLBCL).
